Abstract
Bottom-up proteomics technique is a usual way to identify different proteins in the insect’s world. Digestive system of the insects consists of the salivary glands and alimentary canal. The study of insect digestive system particularly Sunn pest has a key role for finding new potential targets in enzyme-inhibitor interaction. In general, any disruption in the normal process of insect nutrition can reduce or suppress feeding and assimilation in the insects. In the present research, proteome patterns of some of organs in the adult and fifth instar nymph of Sunn pest were determined using 2-dimensional electrophoresis. Sunn pest adults were collected from wheat farms in Torbat-e-Jam, Iran. After 2-days feeding, twenty adults (10 male and 10 female) were selected for dissection. Midgut proteins were visualized by two dimensional electrophoresis patterns. There were as many as 671 spots in the proteome patterns that needed to be identified. Then, a number of spots that changed after feeding were selected as target proteins. These spots were cut and digested with trypsin and then injected to liquid chromatography and mass spectrometry (LC / MS-MS). Afterwards, resulted spectrums were analyzed using information obtained from National Center of Biotechnology with two entries as animals and wheat. Results showed that -amylse, different proteases, catalase, enolase and glyceraldehis3-phosphate were up regulated in the gut of fed Sunn pest in comparison to unfed adults.This result suggests that protein expression in the gut of Sunn pest is affected with feeding directly. In addition to identification of some gut proteins of Sunn pest, this study showed the amount of changed proteins in the digestive system before and after feeding.
